Will Abt Jan 1599/00 - A Chancery Court case, in which the bill is dated 21 Nov 1617 (C 2/JasI/F5/8) mentions this will:
"The said John Glandfell ... did by his last Will & Testament in writing made in or about the moneth of Januarye in the yeare of our Lord god accordinge ot the Computacon of the Church of England One Thousand Five hundred Fowerscore & Nynteene will & devise unto Joane then his wyef & nowe the wyef of your said said Orator all the said landes in these wordes followinge vizt: Item I give & bequeath unto Joane my wyef during her naturall lyef whome I doe make my whole & sole Executrix of this my last will & testamt All my landes & Tenemtes wherein I have any Righte or estate by any manner of waye and shortly after the said John Glandfeile died"
Died Abt Jan 1599/00 - Living when his brother wrote his will and when commission issued to him regarding execution of the will, but died by 23 May 1601 when administartion was granted to his relict.
A Chancery Court case (C 2/JasI/F5/8) describes him as having written his will in about Jan 1599/1600 and died shortly after.

Notes - Son and heir of Matthew (according to Chancery case, C 2/Eliz/W11/11); also described as his eldest brother in the will of Henry.
In his will Henry Glanville refers to his brother John AND to John of Totnes, but unfortunately it is not clear if both were the same person. From the PRs for Totnes I think John-the-Totnes-merchant died in 1634/5, while I think John-the-brother died by 1601, so I think they were two different people.
From a comment in Henry's will, then, if I am right that they were 2 different people, I don't think they can have been brothers. Henry said 'the foresaide John Glandfeilde my brother' - this would have been ambiguous if both were brothers.

Father Matthew GLANVILLE, b. Est 1520, d. Abt 1582 (Age ~ 62 years) Married Aft 1545 - I am guessing if Matthew had been married by 1545 he would be in the lay subsidy listing for that year, but he is not found there.
Some examples I have seen of a person giving two sons the same name they were clearly sons with different mothers. I don't know if this was usually the case, nor whether it applied to daughters as well.
If it was usual then it would seem probable that Matthew married twice. (Even if not indicated by having two daughters Joanne, it was quite likely at this time).
In the absence of any information about wive(s) I am showing all the children as of one marriage.
